A full day is tight but doable. Start at the lower gate at opening time, walk up the travertines (1 h), visit Hierapolis (2-3 h including museum), swim in Cleopatra's Pool (1 h), lunch in the village, back down the travertines at sunset. Overnight stay strongly recommended for less rush.
